Do Re Mi mengisahkan tiga sahabat Masdur/Do (Tan Sri P Ramlee), Amore/Re (A R Tompel) dan Minami/Mi (Ibrahim Din) berjuang demi kehidupan. Do, banyak berhutang, sehingga sanggup menipu kematian dirinya dan isteri, yang terbongkar dan akhirnya menyebabkan Do bercerai dengan isterinya. Do ke bandar memulakan hidup baru. Re pula mempunyai seorang isteri yang bekerja di kelab malam, sementara dia sendiri menguruskan hal rumahtangga. Akibat kecuaian Re, wang isterinya dicuri, lalu Re dihalau keluar dari rumah dan membawa diri mencari kerja di kota.

Mi, seorang bujang yang meminati gadis yang tinggal berhadapan rumahnya, iaitu Milah (Wan Chik Daud). Ketika Mi keluar mencari pekerjaan, dia menolong mengejar pencuri yang merampas beg tangan seorang wanita. Do dan Re yang sedang mencari kerja di pejabat buruh mendengar jeritan wanita tersebut, lalu mengejar Mi yang sedang berlari, kerana menyangka dia adalah pencuri itu. Maka semenjak itu, mereka bertiga berkawan rapat dan berlaku pelbagai perkara lucu. Pelbagai cabaran tiga sahabat dalam mencari ikhtiar hidup.

DO RE MI is the first sequel to the films, followed by Nasib Do Re Mi and Laksamana Do Re Mi. Do Re Mi tells the story of three friends Masdur/Do (P Ramlee), Amore/Re (A R Tompel) and Minami/Mi (Ibrahim Din) wanted to improve their life. Do, heavily indebted, is so willing to cheat on the death of himself and his wife, which is exposed and eventually causes Do to divorce his wife. Do then went to the city to start a new life. Re, on the other hand, has a wife who works in a nightclub, while he himself manages the household affairs. As a result of Re's negligence, his wife's money was stolen, then Re was kicked out of the house and took himself to find work in the city.

Mi, a bachelor who is interested in the girl who lives in front of his house, namely Milah (Wan Chik Daud). When Mi goes out looking for a job, she helps chase down a thief who snatches a woman’s handbag. Do and Re who were looking for work in the labor office heard the woman's screams, then chased after Mi who was running, thinking he was the thief. So since then, the three of them have become close friends and various funny things have happened. Various challenges of three friends in finding a way to support their basic needs.
